5-Amino-1MQ (5-Amino-1-methylquinolinium) is a small-molecule NNMT inhibitor marketed for fat loss and longevity. Despite often being grouped with peptides, it's NOT a peptide — it's a methylquinolinium analog. This distinction matters for access.
Mechanism: 5-Amino-1MQ selectively inhibits NNMT (nicotinamide N-methyltransferase) in adipose tissue. NNMT inhibition reduces the methyl-group sink, preserving NAD+ and SAM pools, which in turn enhances sirtuin activity (SIRT1), improves mitochondrial function, increases fat oxidation, and reduces white adipose tissue in preclinical models.
Clinical evidence: No human clinical trials have been published. All dosing is extrapolated from preclinical rodent data (Kannt et al. 2018, Nat Chem Biol).
Standard protocol: 50–150 mg oral daily, once in the morning or pre-workout. Typical titration: 50 mg x 1–2 weeks → 100 mg daily (most common maintenance) → 150 mg (upper end; plateau reported beyond this). Cycles are 8–12 weeks on, 4–8 weeks off.
Forms: Oral capsules dominate (50 mg is the standard strength, typically 60 per bottle). Lyophilized powder is also sold for research but requires self-encapsulation.
Regulatory status (April 2026): NOT FDA-approved.
